A single TEM image is a 2D projection of a 3D object - it doesn’t provide any depth information.

In a typical Transmission Electron Microscopy (i.e., TEM) setup, images are taken at one to two degree rotation intervals.

Gold particles and cross-correlation algorithms are then used to align individual images. Image reconstruction typically happens using back-projection.
